Abstract

Liver disorders remain a major global health burden, and conventional hepatoprotective drugs are limited by cost, availability, and side effects, driving renewed interest in polyherbal formulations. This study evaluates the pharmacological rationale and evidence base for a polyherbal formulation combining Tinospora cordifolia, Boerhavia diffusa, Moringa oleifera, and Phyllanthus emblica — four plants independently reported to possess hepatoprotective, antioxidant, and anti-inflammatory properties. The research problem addressed is the absence of a consolidated pharmacological evaluation framework for this specific four-plant combination, despite strong individual evidence for each constituent. A structured evaluation protocol was designed using the carbon-tetrachloride (CCl₄)-induced hepatotoxicity model in Wistar rats, with biochemical liver markers (SGPT, SGOT, ALP, total bilirubin), antioxidant enzyme assays, and histopathological scoring as primary endpoints, alongside standard statistical evaluation (one-way ANOVA with Dunnett's post-hoc test). The evaluation draws on convergent literature evidence indicating that individual and related polyherbal combinations of these plant families typically produce 40–70% restoration of elevated liver enzymes toward normal values, with dose-dependent antioxidant enhancement. The formulation's proposed mechanism operates through free-radical scavenging, membrane stabilization, and hepatocyte regeneration support. The study concludes that the four-plant combination has strong literature-supported pharmacological rationale and proposes a validated, reproducible protocol for future experimental confirmation. This work contributes a consolidated evidence synthesis, a standardized evaluation design, and quantitative benchmarking equations that can guide subsequent in vivo trials of this and related polyherbal hepatoprotective formulations.
