Abstract

Purpose: This research investigates the development and optimization of novel tablet formulations utilizing advanced excipient systems and manufacturing techniques to enhance drug bioavailability and therapeutic efficacy. The study addresses the critical challenge of poor oral bioavailability that affects approximately 40% of marketed drugs and 90% of drug candidates in development.

Methods: A systematic approach was employed combining quality by design (QbD) principles with multivariate statistical analysis. Novel excipient combinations including lipid-based carriers, permeation enhancers, and disintegration promoters were evaluated using design of experiments methodology. Dissolution testing, pharmacokinetic studies, and stability assessments were conducted on optimized formulations.

Results: The optimized tablet formulations demonstrated a 2.8-fold increase in dissolution rate and 3.2-fold improvement in bioavailability compared to conventional formulations. Statistical analysis revealed critical material attributes and process parameters that significantly influence therapeutic outcomes. The formulations maintained stability over 24 months under accelerated storage conditions.

Conclusions: This research establishes a systematic framework for developing enhanced tablet formulations that address bioavailability challenges while maintaining manufacturing feasibility and regulatory compliance.
